You changed the path to an incorrect one.
I take it you saw a dialog box before this error that asked you to locate the Fable directory?
Essentially, the application for some reason wasn't able to create the Fable SetupPath registry variable.
The FableRegistry variable in the INI file should not be changed, in most situations. It should be:
FableRegistry = Software\Microsoft\Microsoft Games\Fable\1.0
There are some versions of the game that have a slightly different reg key, for whatever reason.
The MyPath one can be changed.

My setupPath (in the registry) is D:\Fable - The Lost Chapters\
in the CBox.ini, I have
FableRegistry = Software\Microsoft\Microsoft Games\Fable\1.0
MyPath = D:\Fable - The Lost Chapters\
I have try with the ";", without it, changing the registry key, adding or removing some "\"...
Now I have :
EventType: clr20r3
